Abstract:
The subcellular distribution of cargoes is crucial for cell fate and therapy. However, precise delivery to specific intracellular targets remains challenging. Recently, we developed a novel ultra-pH-sensitive (UPS) nanotechnology based on the cooperative self-assembly of ionizable block copolymers. We applied this to create a combinatorial library of Specific ORganelle-Targeting nanomedicines (nanoSORT) composed of three modules: (i) a stimuli-responsive module sensing and remodeling the tumor microenvironment; (ii) on-demand release module for precise payload delivery; (iii) targeted drug module for specific organelle delivery. This design enables delivery to multiplex organelles, such as early endosomes. Using nanoSORT, we achieved spatiotemporal targeting of different stages of endosomal maturation, enabling tunable cellular pyroptosis. Through organelle cascade targeting, payloads are selectively sorted into mitochondria, inducing reactive oxygen species stress there to enhance pyroptosis-based cancer immunotherapy. This nanotechnology provides a promising platform for nanomedicine research and clinical translation.

Prof. Yiguang Wang is a Distinguished Professor at Peking University School of Pharmaceutical Sciences and serves as the vice dean for research and academic affairs. His research focuses on smart nanomedicine for disease diagnosis and treatment. He has authored over 70 papers in prestigious journals, including Nat. Mater., Nat. Nanotechnol., Nat. Commun., J. Am. Chem. Soc., etc. Prof. Wang has been funded with prestigious grants including the National Science Fund for Distinguished Young Scholars, the National Science Fund for Excellent Young Scholars, and the Beijing Science Fund for Distinguished Young Scholars. He has been honored with the Youyou Tu Youth Scholar Award, the Bayer Investigator Award.
