Abstract:
This speech presents an innovative approach to enhance electron transfer-based therapies by coupling electroactive liposome membranes with nanoparticles for cancer treatment and cellular imaging applications. Leveraging the extracellular electron transfer capabilities of Shewanella oneidensis MR-1, membrane-integrated liposomes (MILs) rich in c-type cytochromes (c-Cyts) were developed using a liposome fusion-induced membrane exchange method. These electroactive MILs improve the efficacy of nanoparticles such as TiO2, lanthanide-based upconversion nanoparticles (UCNPs), and gold nanoparticles (Au NPs). First, TiO2@MIL nanoparticles demonstrated enhanced superoxide anion production and hydroxide radical generation under low-dose X-ray irradiation, indicating significant potential for radiocatalytic cancer treatments. Second, UCNP@MIL constructs exhibited enhanced upconversion efficiency under near-infrared excitation by facilitating electron transfer from the electroactive membrane to the UCNP core, bypassing the ligand-sensitized mechanisms. Lastly, Au@MIL constructs disrupted the redox balance in cancer cells by serving as electron sinks, leading to oxidative stress, lipid oxidation, and apoptosis under hypoxic conditions without relying on iron-mediated pathways. These findings demonstrate the versatility of electroactive MILs in advancing nanoparticle-based therapies and imaging, offering promising new avenues for cancer treatment and biological applications.
Bio:
Prof. Chen-Sheng Yeh is Chair Professor in the Department of Chemistry, National Cheng Kung University. He was awarded the excellent research awards by Taiwan’s Ministry of Science and Technology in 2010 and 2016 and the Chair Professorship from Taiwan’s Foundation for the Advancement of Outstanding Scholarship in 2016. He is also a recipient of the K.T. Li Honorary Scholar Award, National Cheng Kung University in 2017, and the excellent technology transfer award from Taiwan’s Ministry of Science and Technology in 2018. His research focuses on functional nanomaterials for biological applications and nanostructured characteristics. He has published over 130 SCI papers with a Google Scholar h-index of 70.
